Tento excitovan\u00fd stav je energeticky nestabiln\u00ed. Elektrony se rychle vracej\u00ed do z\u00e1kladn\u00edho stavu a p\u0159i tom uvol\u0148uj\u00ed absorbovanou energii. Fotodioda m\u011b\u0159\u00ed intenzitu z\u00e1\u0159en\u00ed emitovan\u00e9ho fluorescenc\u00ed p\u0159i ur\u010den\u00e9 vlnov\u00e9 d\u00e9lce v modr\u00e9m sv\u011bteln\u00e9m rozsahu. Intenzita se m\u011b\u0159\u00ed v RFU (Relativn\u00ed fluorescen\u010dn\u00ed jednotky). \u010c\u00edm vy\u0161\u0161\u00ed je hodnota RFU, t\u00edm vy\u0161\u0161\u00ed je intenzita fluorescen\u010dn\u00edho sv\u011btla.   <\/p>\n\n\n\n<p class=\"wp-block-paragraph\">D\u00edky kalibraci se standardy fluorescence SITA jsou nam\u011b\u0159en\u00e9 hodnoty RFU srovnateln\u00e9.<\/p>\n\n\n\n<p class=\"wp-block-paragraph\">Fluorescen\u010dn\u00ed m\u011b\u0159ic\u00ed za\u0159\u00edzen\u00ed SITA pracuj\u00ed na principu konfok\u00e1ln\u00edho m\u011b\u0159en\u00ed. Excita\u010dn\u00ed sv\u011btlo a sv\u011btlo emitovan\u00e9 fluorescenc\u00ed sleduj\u00ed stejnou paraleln\u00ed dr\u00e1hu. Fluorescence m\u011b\u0159en\u00e1 v kapalin\u011b nebo na povr\u0161\u00edch je kumulativn\u00edm parametrem v\u0161ech p\u0159\u00edtomn\u00fdch fluorescen\u010dn\u00edch l\u00e1tek. Kyseliny karboxylov\u00e9, jejich estery a alifatick\u00e9 ketony tak\u00e9 fluoreskuj\u00ed.  <\/p>\n\n\n\n<p class=\"wp-block-paragraph\">M\u011b\u0159en\u00ed fluorescence dok\u00e1\u017ee zjistit i nejmen\u0161\u00ed mno\u017estv\u00ed fluorescen\u010dn\u00edch l\u00e1tek. Dokonce i otisky prst\u016f, kter\u00e9 obsahuj\u00ed m\u00e9n\u011b ne\u017e 2 % ko\u017en\u00edch tuk\u016f, mohou b\u00fdt detekov\u00e1ny. Typick\u00e9 rozsahy detekce pro v\u00fdrobn\u00ed pom\u016fcky jsou 10 mg\/m\u00b2 \u2026 10 g\/m\u00b2 (1 \u03bcg\/cm\u00b2 \u2026 1 mg\/cm\u00b2).  <\/p>\n\n\n\n<p class=\"wp-block-paragraph\">V\u00fdjimkami jsou l\u00e1tky, kter\u00e9 nefluoreskuj\u00ed p\u0159i UV excitaci, jako jsou silikonov\u00e9 oleje, nasycen\u00e9 organick\u00e9 slou\u010deniny, kr\u00e1tko\u0159et\u011bzcov\u00e9 nenasycen\u00e9 uhlovod\u00edky, kovy a jejich oxidy. Zda je mo\u017en\u00e9 detekovat kontaminaci v mno\u017estv\u00ed relevantn\u00edm pro proces (lepen\u00ed, sva\u0159ov\u00e1n\u00ed apod.), lze otestovat jednoduch\u00fdm fluorescen\u010dn\u00edm testem. <\/p>\n\n\n\n<p class=\"wp-block-paragraph\">P\u0159id\u00e1n\u00edm fluorescen\u010dn\u00edch pigment\u016f nebo barviv jako fluorescen\u010dn\u00edch marker\u016f mohou b\u00fdt detekov\u00e1ny i slab\u011b nebo nefluoreskuj\u00edc\u00ed l\u00e1tky.
